Deadlifts will get your lower back, so it's might complement your butt , but won't add anything to your butt itself.
 
definitely the squat. work your quads and hams before hand so that your glutes can do most of the work.

oh and, use lotion or something generously so that you dont get stretch marks on your big booty.
 
Remove of body fat, you probarly got some muscles there unless you sit in front of the computer 24/7.
 
Squats, lunges and cardio. I guess the amount and whatnot would depend on your body in it's current state. Do you go to the gym at all currently or just starting up?
 
i think you guys need to read up on dead lifts pronto....as they do much more than your lower back
